德國美因茨 & 美國馬里蘭州羅克維爾，2023年6月2日（GLOBE NEWSWIRE）——BioNTech SE（納斯達克：BNTX）與 OncoC4, Inc. 今日公佈了抗CTLA-4抗體候選藥物BNT316/ONC-392（gotistobart）在轉移性、PD-(L)1 耐藥非小細胞肺癌（NSCLC）患者中的積極初步資料。結果顯示，該藥單藥即可在這一難治的患者人群中產生臨床獲益，且耐受性良好。資料將以壁報討論形式在2023年美國臨床腫瘤學會（ASCO）年會展示，報告人為美國俄亥俄州立大學 Pelotonia免疫腫瘤學研究所 Kai He 博士。

本次 ASCO 發佈的結果共納入27例至少接受兩次10 mg/kg BNT316/ONC-392 給藥、可評估腫瘤反應的轉移性 PD-(L)1 耐藥 NSCLC 患者。資料截止時，客觀緩解率（ORR）為 29.6%，疾病控制率（DCR）達 70.4%，其中 1 例完全緩解、7 例部分緩解、11 例疾病穩定。3–4 級免疫相關不良事件（irAE）見於 10 例患者（30%），低於同類藥物歷史報導。

肺癌仍是全球最常見、最致命的惡性腫瘤，其中NSCLC占比約 85%。基於上述資料，雙方計畫於2023年第三季度啟動單藥III期註冊研究 PRESERVE-003（NCT05671510），該研究已獲FDA快速通道資格。

關於BNT316/ONC-392

BNT316/ONC-392（gotistobart）為BioNTech與OncoC4聯合開發的下一代抗CTLA-4抗體，正處晚期臨床階段。其設計保留 CTLA-4迴圈機制，維持外周組織 Treg 功能，以期降低免疫相關毒性、提升安全性。目前正在進行 I/II 期試驗（NCT04140526）評估其單藥或與 pembrolizumab 聯合用於晚期實體瘤；另有一項 II期研究（NCT05446298）探索與 pembrolizumab 聯用治療鉑耐藥卵巢癌。

關於非小細胞肺癌（NSCLC）

非小細胞肺癌（NSCLC）涵蓋除小細胞肺癌外的所有上皮性肺癌，包括鱗癌、大細胞癌及腺癌，占肺癌病例約 85%。美國 2012–2018 年五年相對生存率僅 23%，治療選擇有限，現行標準包括手術、放療及化療。
