德國美因茨 & 美國馬里蘭州羅克維爾，2023年3月20日 - BioNTech SE（納斯達克：BNTX，“BioNTech”）與專注于發現和開發癌症治療新藥的臨床階段生物製藥公司OncoC4, Inc.今日宣佈，雙方已簽署獨家全球許可與合作協定，共同開發和商業化OncoC4的下一代抗CTLA-4單克隆抗體候選藥物 ONC-392，涵蓋單藥或聯合療法用於多種癌症適應症。交易預計於2023年上半年完成，須滿足慣例交割條件及監管批准。

CTLA-4 是一種通過多種機制抑制免疫細胞活性的分子。OncoC4的CTLA-4 抗體候選藥物ONC-392旨在清除腫瘤微環境中的免疫抑制性T細胞（Tregs），同時保留健康組織中的Tregs。憑藉潛在差異化的安全性特徵，ONC-392有望在臨床實現更有效給藥方案並提升腫瘤殺傷效果。2021及2022年SITC大會公佈的正在進行中的I/II期研究（NCT04140526）資料顯示，ONC-392單藥或與pembrolizumab聯合用於晚期實體瘤患者——尤其是已進展於PD-1和CTLA-4免疫治療的患者——展現出令人鼓舞的臨床活性。

ONC-392 已獲美國食品藥品監督管理局（FDA）授予“快速通道”資格，作為免疫治療耐藥非小細胞肺癌（NSCLC）的單藥治療。針對 PD-1 耐藥 NSCLC 的單藥資料支援啟動隨機 III 期研究（NCT05671510），將 ONC-392 單藥與當前標準治療進行對照。該候選藥物還在另一項 II 期研究中與 pembrolizumab 聯合用於鉑耐藥的卵巢癌患者（NCT05446298）。

根據協定條款，OncoC4 將獲得 2億美元首付款，並有資格獲得後續開發、監管及商業里程碑付款，以及雙位數階梯式特許權使用費。BioNTech 與 OncoC4 將共同推進 ONC-392 單藥及與抗 PD-(L)-1 抗體聯合用於多種實體瘤適應症（包括 NSCLC）直至獲批，相關研究費用由雙方均等分擔。所有超出 PD-1 抑制範疇的聯合方案，特別是與 BioNTech 自有研發管線的聯用，將由 BioNTech 獨立開發。BioNTech 擁有上述任何產品的獨家全球商業化權利，未來將在部分市場與 OncoC4 共同推廣，具體細節另行商議。

ONC-392是OncoC4開發的下一代抗CTLA-4抗體候選藥物。免疫檢查點受體 CTLA-4可抑制 T細胞免疫應答，降低其識別並清除癌細胞的能力。阻斷 CTLA-4 可維持 T細胞活性並增強抗腫瘤效應。ONC-392在設計上有意保留 CTLA-4 的迴圈機制，從而維持外周組織中 Treg的功能，旨在減少免疫相關不良反應並提升安全性。
