德国美因茨 & 美国马里兰州罗克维尔，2023年6月2日（GLOBE NEWSWIRE）——BioNTech SE（纳斯达克：BNTX）与 OncoC4, Inc. 今日公布了抗CTLA-4抗体候选药物BNT316/ONC-392（gotistobart）在转移性、PD-(L)1 耐药非小细胞肺癌（NSCLC）患者中的积极初步数据。结果显示，该药单药即可在这一难治的患者人群中产生临床获益，且耐受性良好。数据将以墙报讨论形式在2023年美国临床肿瘤学会（ASCO）年会展示，报告人为美国俄亥俄州立大学 Pelotonia免疫肿瘤学研究所 Kai He 博士。
本次 ASCO 发布的结果共纳入27例至少接受两次10 mg/kg BNT316/ONC-392 给药、可评估肿瘤反应的转移性 PD-(L)1 耐药 NSCLC 患者。数据截止时，客观缓解率（ORR）为 29.6%，疾病控制率（DCR）达 70.4%，其中 1 例完全缓解、7 例部分缓解、11 例疾病稳定。3–4 级免疫相关不良事件（irAE）见于 10 例患者（30%），低于同类药物历史报道。

关于BNT316/ONC-392
BNT316/ONC-392（gotistobart）为BioNTech与OncoC4联合开发的下一代抗CTLA-4抗体，正处晚期临床阶段。其设计保留 CTLA-4循环机制，维持外周组织 Treg 功能，以期降低免疫相关毒性、提升安全性。目前正在进行 I/II 期试验（NCT04140526）评估其单药或与 pembrolizumab 联合用于晚期实体瘤；另有一项 II期研究（NCT05446298）探索与 pembrolizumab 联用治疗铂耐药卵巢癌。
